The explosion late on Wednesday, happened after a bomb, planted by unknown terrorists, exploded when the vehicle passed by it.
A conscript was also injured in the explosion, the source said.
After the incident, the army and police forces closed the road with checkpoints in search for the terrorists.
In a separate incident, security forces killed two militants, belonging to Ansar Beit el-Maqdes terror group, late on Wednesday in Abu Regila area in Al-Salam district in Cairo.
The exchange of fire left two militants dead and three civilians and two policemen were injured in it.
The forces are currently searching for the third militant who escaped during the clashes.
Militant attacks targeting police and military have increased after the ouster of Islamist ex-president Mohamed Morsi last year. Over 500 security personnel have been reported killed since then.
