The 64-year-old actor will be seen on the April 1 episode "Inside Man" of the 10th season, reported Variety.
The episode will see show Sam and Castiel, who needed help from someone in heaven, seeking out Bobby.
Bobby died in season seven's "Death's Door", which happens to be lead actor Jensen Ackles' favourite episode.
He last appeared in the season 9 premire, though was referenced heavily in this season's sixth episode, "Ask Jeeves".
The character has been a beloved staple of the series since his appearance in the season 1 finale.
