Holmes has signed on to play Lorna in Theresa Rebeck's new play, Dead Accounts. The five-character comedy is set to open later this year at the Music Box Theatre and will be directed by three-time Tony Award winner Jack O'Brien.
It is Holmes first major career move since she surprised Hollywood by filing for divorce from Cruise a few weeks ago.
The actress has already filmed an episode of 'Project Runway'. She is said to be working on the indie film Molly, which she also co-wrote and produced.
Holmes had garnered mixed reaction for her Broadway debut in 2008's 'All My Sons'.
