The 2006 X Factor runner-up married Emma Stephens in 2012 after meeting when they both starred in a West End production of 'Grease' four years earlier, reported Digital Spy.
Quinn, who confirmed the news in a statement, has vowed to continue with his UK tour and will still release his new single as planned.
He said: "Unfortunately the speculation is true and Emma and I have regrettably parted ways.
"Out of respect for the privacy of my family I don't want to delve into my private life too much however what I will say is that our lives, after a period of time, have gone down separate paths."
Quinn recently released two versions of 'The Beauty of Who You Are' - a ballad and a dance track - to raise money for Parkinson's UK.
