Astin, 29, said the "Pitch Perfect 3" will not include the franchise's main all-boy a cappella group as the makers are planning to change the plot.
"Yes. As of now, the Trebles and I won't be in the third Pitch Perfect movie. They seem to be taking the story in a different direction," he wrote.
Astin's Jesse, along with fellow Treblemakers Bumper (Adam Devine) and Benji (Ben Platt), continually competed against Barden University's all-female a cappella group the Bellas, among others, through the films.
"Orange is the New Black" actress Ruby Rose also recently joined the film, which will be directed by Trish Sie, the helmer of "Step Up All In".
It is set to hit the US theaters on December 22, next year.
