Lyari Superintendent Police Aftab Nizamani was patrolling with his guards when the gunmen opened fire on his car, officials said.
However, the police officer escaped unhurt as his security guards fired back killing the two gunmen. The two attackers are yet to be identified.
Lyari has become notorious for gang wars with criminals carrying out a reign of terror in the low income area which remains the stronghold of the Pakistan Peoples Party which rules the southern Sindh province.
In the past, senior police officials, paramilitary rangers and even traffic police officials have come under attack in Karachi where the federal government is carrying out a clean-up operation against militancy, target killings and sectarian violence.
