The car, on its way to Shillong from Manipur, was stopped by a home guard as it was driving on the wrong side of the road, police said.
Two people immediately fled the scene while the third was nabbed by the police, district SP Davis Marak said.
"The incident took place around 7 am when an SUV with a Manipur registration number plate was stopped by a home guard near a petrol pump at Mawblei for violating traffic rules," he said.
According to an official of the anti-narcotics division, the suspected smugglers had sourced the contraband from Moreh along the India-Myanmar border.
"We are conducting a search to nab the other two accused in the case," Marak added.
