Clooney, 53, is set to tie the knot with human rights lawyer fiancee Amal Alamuddin, 36, this weekend in Venice, Italy, reported Us magazine.
Alessandro Greco, whom Clooney calls Sandro, will apparently take a day off to attend the wedding among such A-list guests as Matt Damon, Sandra Bullock, Anna Wintour, and Emily Blunt.
Sandro has been driving the 'Gravity' actor for years, and was on hand to charter Clooney and his friends on Friday, September 26, to their bachelor's dinner.
Taking over Sandro's spot for the day will be his assistant, whose first job will be to pick up the Oscar winner for his big day.
