The 55-year-old actor has closed a deal to star on the nine-episode limited series, reported Ace Showbiz.
MacLachlan made a surprise appearance at the Television Critics Association's winter press tour.
"I'm very excited to return to the strange and wonderful world of 'Twin Peaks'. May the forest be with you," he said.
The 'Twin Peaks' limited series is eyed to air in 2016. Series co-creator David Lynch will direct all nine episodes.
The limited series is set in the present day and is promised to "continue the lore of the original series."
