South Africa dealt with a massive blow as fast bowler Dale Steyn left the field in the middle of the over following a strain on his right shoulder.
Dane Piedt, however, provided some resistance to the hosts as he dismissed skipper Alastair Cook and opener Alex Hales for their respective scores of seven and 26, Sport24 reported.
At the time of tea, Nick Compton (15) and Joe Root (six) were at the crease.
Earlier, Stuart Broad and Moeen Ali bagged four wickets each as South Africa were bundled out for 214 runs in their chase of England's first inning score of 303.
