The movie will be directed by "Boyhood" helmer Richard Linklater for Annapurna Pictures, said The Hollywood Reporter.
The novel has been adapted by "Fault in Our Stars" writers Scott Neustadter and Michael H Weber.
It centres on an agoraphobic architect and mother named Bernadette Branch who goes missing prior to a family trip to Antarctica. The book is narrated by her 15-year-old daughter Bee Branch.
The novel unfolds in its own quirky way - via emails, letters, FBI documents, correspondence with a psychiatrist and a bill for an emergency room visit involving the titular character.
Earlier this year, Blanchett, 46, starred as the wicked stepmother in Disney's live-action Cinderella.
