Martin jumped in to replace Emma Stone during a crucial kiss scene which the pair struggle to get right, reported Digital Spy.
Demanding a wig and holding Garfield in his arms, Martin said, "Now you listen to me, you beautiful spidery man. With great power comes great responsibility, and our responsibility right now is to make sweet, sweet love.
"So, why don't you go ahead and show me your beautiful tongue?"
Garfield hosted the NBC sketch comedy series for the first time in his career, following a brief cameo on the show in 2011 fronted by Stone.
