Clinton has become the first female presidential nominee of Democratic party.
"What does it take to be the first female anything? It takes grit and it takes grace," Streep, 67, said as she took to stage.
In a moving speech, the actress placed Clinton, who is 68, among the pioneering American women, from Amelia Earhart and Harriet Tubman to Sandra Day O'Connor, Deborah Sampson and Geraldine Ferraro.
Streep told the crowd that they had made history by selecting Clinton, who will make a great first female president.
"Nearly 100 years after women got the vote, you people have made history. And you're going to make history again in November because Hillary Clinton will be our first woman president, and she will be a great president.
