Police said the deceased men, hailing from Palayamkottai in tirunelveli district, were activists of Ulaga Tamizhar Vazhvumirai Katchi.
They were returning after participating in the party's conference at Thanjavur when the mishap occurred.
Police suspect that one of the activists, who was driving the car, might have dozed off, leading to the mishap.
