The film is a story about a renegade Russian military leader whose actions lead to the brink of war with the US, reported Deadline.
Dafoe has been cast as Russian submarine Captain Andropoyov.
Martin Campbell is set to direct the film, which is based on the novel 'Firing Point' by George Wallace and Don Keith.
"Hunter Killer" is scheduled to start filming in New Orleans in July.
Dafoe will next be seen alongside Matt Damon in "The Great Wall", directed by Zhang Yimou.
