The 50-year-old actor, who is currently working on the television series "Kingdom", will begin shooting the film in September, reported Entertainment Weekly.
Grillo played Sergeant in the 2014 "The Purge: Anarchy", a man who seeks revenge on the night of the Purge, only to get pulled into protecting a group of people who are left without shelter as purging begins.
The latest sequel will be written and directed by "The Purge" franchise creator James DeMonaco.
The film will take place in the future where one day, each year, all crime is legalised for a 12-hour period, described by the government as "The Purge."
"The Purge 3" is expected to release in July next year.
